| Title | ANALYSIS THE TEAR PROTEOME OF SJOGREN'S SYNDROME PATIENTS WITH TIMSTOF PRO |
| Purpose | To investigate differentially modulated tear proteins (DMTPs) of Sjögren's syndrome patients (SSp) using a highly sensitive trapped ion-mobility quadrupole time-of-flight mass spectrometry (timsTOF Pro). |
| Methods | Tear samples were collected from 6 healthy controls (HC) and 6 SSp using Schirmer strips. The protein content of each sample was normalized to 250 ng. Proteomics analysis was performed with timsTOF Pro and data was processed with MaxQuant software for protein identification. |
| Results | In HC and SSp 569 proteins were common, but 65 proteins were unique in SSp and 264 in HC. In SSp 65 proteins were up-regulated and 110 down-regulated with a fold-change ≥ 1.5 (p-value ≤0.05). Catalytic activity (46.9%) and binding (38.5%) formed the main groups of molecular function among significantly modulated proteins. Binding proteins were mainly down-regulated (38 versus 17). A large number of oxidoreductase enzymes such as Peroxiredoxin-1,-5,-6, sulfhydryl oxidase-1, glutathione peroxidase-3 and lactoperoxidase also were down-regulated. Inversely, proteins involved in apoptosis such as caspase-3 and proteasome subunits were upregulated and glutathione synthase and Calmoduline3 related to biological metabolism were up-regulated in SSp. These results highlight an alteration in cellular process and antioxidant activity.

| Conclusion | DMTPs that involve in important signaling pathways such as oxidative stress and cell death were revealed in SSp. The improved spatial resolution, high sensitivity, speed, and specificity of timsTOF Pro can foster the investigation of candidate biomarkers to elucidate the pathophysiology of SS. |
